The first synthetic gene expression system for the tetracycline-responsive promoter of the human gene for human fucosyltransferase, has been developed, and its expression in the cells of human choriomastatycosis (CH) is modified by a tetracycline-inducible gene expression system. We have used the tetracycline-inducible gene expression system to develop an expression vector, which will be described here. Our strategy is to add a tetracycline-responsive promoter to the genome of choriomastatycosis, and to use the tetracycline-inducible gene expression system for the expression of this gene in a single cell culture. The tetracycline-inducible promoter is used to express fucosyltransferase from the plasmid of the plasmid pPRG-TetO-tetO-Fuc, which can be expressed from the plasmid pPRG-TetO-TetO. A tetracycline-inducible gene expression system for choriomastatycosis was created by using the pPRG-TetO-TetO-Fuc plasmid. The pPRG-TetO-TetO-Fuc vector was then injected into a single cell culture of a choriomastatycosis patient.
This research has developed a novel vector for the production of tetracycline-inducible gene expression plasmids, which will be described herein. The vector will be obtained fromShandong Z. Jing(Shanghai) andZhejiang Z. Zhang(Zhejiang), and it will be constructed by inserting the pPRG-TetO-TetO-Fuc plasmid into the pPRG-TetO-TetO-Fuc vector. A series of bacterial strains will be used to test the expression of the tetracycline-inducible gene expression plasmid in the cells of the patient.
The tetracycline-responsive promoter of the human gene for human fucosyltransferase is a modified version of the previously characterized promoter of a gene encoding for the gene of interest (GOI).
